Beyond tau and TREM2: checkpoint inhibition, inflammasome drugs, brain delivery technologies, and biomarker-led diagnostics reshape Alzheimer's pipeline.
The first article in this series examined how Alzheimer’s drug development is moving beyond amyloid towards tau and microglial biology. Tau-lowering approaches aim to address the pathology most closely associated with clinical progression, while TREM2 agonists seek to improve the brain’s response to plaques and neuronal injury. What is becoming more evident is that new therapies must act at the right disease stage, reach the right tissue and deliver a clinical benefit that justifies their risks and treatment burden. The development pipeline is aptly now extending as such into peripheral immunity, chronic inflammation and cellular stress, supported by advances in brain delivery and biomarker-led diagnosis.
