Simcere Zaiming Licenses Sim0660 to Roche for $1.53 billion

Simcere Zaiming has licensed its first-in-class tri-specific CD79a/CD19/CD3 T-cell engager Sim0660 to Roche for up to $1.53 billion, including a $75 million upfront payment.

Simcere Zaiming, a subsidiary of Simcere Pharmaceutical Group (2096.HK), entered into an exclusive global licensing agreement with Roche for the development and commercialization of Sim0660, a tri-specific antibody targeting CD79a, CD19, and CD3, with therapeutic potential across B-cell-mediated diseases.

Under the terms of the agreement, Roche is set to acquire exclusive global rights to develop, manufacture, and commercialize Sim0660, while Simcere Zaiming is eligible to receive up to $1.53 billion in total payments, including $75 million upfront, along with tiered royalties of up to double digits on future net sales.1

The deal hands Roche a molecule built on a dual-targeting approach that Simcere Zaiming says has not previously reached this stage of development, positioning the companies to pursue a broad set of B-cell-mediated conditions where existing CD20- or CD19-directed therapies have left gaps.1 The payment structure combines a modest upfront $75 million against total potential consideration upwards of $1.53 billion, weighting the bulk of the deal's value toward development, regulatory, and commercial milestones still to be achieved, on top of tiered double-digit royalties on any future net sales once the therapy reaches the market.1

What is Sim0660?

Sim0660 is a tri-specific antibody developed by Simcere Zaiming using its T-cell engager poly-specific antibody technology.1 The molecule combines a CD3-engaging arm with binding domains targeting the two B-cell antigens CD79a and CD19 to induce potent T cell-mediated cytotoxicity while limiting cytokine release. With its potential first-in-class dual-targeting approach to CD79a and CD19, Sim0660 is designed to provide broad coverage across B-cell-mediated diseases and may offer a differentiated therapeutic approach for patients previously treated with CD20- or CD19-directed therapies.1

Why does dual B-cell targeting matter?

Beyond its initial applications, Sim0660 also has therapeutic potential for B-cell-mediated autoimmune diseases, where targeted depletion of pathogenic B cells may provide clinical benefit.1 By pairing CD79a and CD19 binding domains with a CD3-engaging arm in a single molecule, the candidate is designed to broaden coverage across B-cell populations relevant to both malignant and autoimmune disease, while its cytokine-limiting design aims to address a known tolerability challenge for T-cell engager therapies.

What does this mean for Simcere's licensing strategy?

The Roche agreement extends a run of out-licensing activity for Simcere Pharmaceutical Group, which has now completed six such transactions with a potential aggregate total consideration exceeding $6.1 billion. That track record spans multiple deals, including one with Boehringer Ingelheim, built around Simcere Zaiming's proprietary T-cell engager poly-specific antibody technology, the same platform underpinning Sim0660, suggesting the Roche agreement is one output of a broader pipeline strategy built on that technology rather than a standalone transaction.1.2

The deal adds Roche to a growing list of global partners for Simcere's pipeline and gives Simcere Zaiming's T-cell engager platform validation from one of the industry's largest oncology and immunology players, as Mult-specific antibody approaches continue to draw competitive interest.
