BeOne Medicines and Revolution Medicines announced a multi-part collaboration built around a clinical collaboration evaluating drug combinations between select clinical-stage oncology assets from BeOne with any of Revolution Medicines' four clinical RAS(ON) inhibitors.

The deal also includes a separate regional rights agreement granting BeOne exclusive development and commercialization rights to Revolution’s assets in select Asian markets.1

By combining BeOne's oncology pipeline and its established regional research, development, and commercial infrastructure with Revolution Medicines' RAS(ON) inhibitor portfolio, the two companies are positioning themselves to test new combination strategies for RAS-addicted cancers while extending the geographic footprint of Revolution Medicines' therapies into markets where it has not previously operated.1

What will the clinical collaboration focus on?

The clinical collaboration is expected to explore potential targeted combination approaches for patients diagnosed with RAS-addicted cancers. Potential drug combinations for development under the collaboration will draw on certain BeOne assets alongside Revolution Medicines' four clinical RAS(ON) inhibitors:

  • Daraxonrasib: An investigational, oral RAS(ON) multi-selective, non-covalent inhibitor that is not approved by any regulatory authority, including in the U.S. or Europe.2 
  • Elironrasib: A mutant-selective inhibitor that binds selectively and covalently to the oncogenic RAS(ON) form of the RAS G12C variant that drives approximately 12% of cases of NSCLC.3
  • Zoldonrasib: An oral RAS(ON) G12D-selective covalent tri-complex inhibitor.4
  • RMC-5127: An inhibitor that binds to cyclophilin A, creating a complex that selectively recognizes and inhibits the oncogenic RAS(ON) form of the RAS G12V variant.5

Planned combination studies include BeOne's MTA-cooperative PRMT5 inhibitor, BGB-58067, and an EGFR x MET x MET trispecific antibody, BG-T187, each to be studied in combination with either Daraxonrasib or Zoldonrasib.1

What does the regional rights agreement include?

The regional rights agreement leverages BeOne's established research, development, and commercial expertise alongside Revolution Medicines' clinical-stage RAS(ON) inhibitor portfolio.1 Under terms of the agreement, Revolution Medicines has granted BeOne exclusive rights in select Asian markets to develop and commercialize, or in some markets solely commercialize, the same four clinical-stage RAS(ON) inhibitors covered by the clinical collaboration.1

In exchange, Revolution Medicines is eligible to receive development and sales milestone payments, along with tiered royalties on net sales, in the partnered region.1 As part of the deal, Revolution Medicines will retain development and commercial rights to all of its assets outside the licensed territory, including in Japan and South Korea.1

BeOne will also fund and conduct a global registrational Phase III study for one of Revolution Medicines' RAS(ON) inhibitors, using what the company describes as its differentiated, fully in-house development superhighway.1

John V. Oyler, co-founder, chairman, and chief executive officer of BeOne, says the collaboration gives the company a chance to pair its own oncology pipeline with a broad set of RAS(ON) inhibitors while extending its reach through regional infrastructure already in place.
